The genesis of Istanbul University is often linked to the aftermath of the conquest of Istanbul by Fatih Sultan Mehmet in 1453, wherein certain buildings within Ayasofya and Zeyrek were repurposed as medrese (theological schools). Additionally, some scholars attribute the inception to the establishment of Fatih Medrese in 1470 or the post-conquest era.

During the medrese period, Fatih Sultan Mehmet sought to elevate Istanbul as both a political and cultural hub. Scholars summoned to Istanbul post-conquest were appointed to medrese in Zeyrek and Ayasofya, where subjects like theology, law, literature, mathematics, and astronomy were taught.

The founding of Fatih Medrese in 1470 marked a significant stride in medical education. Accompanied by an expansive hospital called Darüşşifa, this institution pioneered medical studies, eventually forming the nucleus of Istanbul University's Faculty of Medicine. The medical education carried out at Darüşşifa endured for about 350 years, contributing substantially to the emergence of Istanbul University's Faculty of Medicine. Medrese institutions remained primary centers of higher education in the Ottoman Empire until the 18th century.

The Tanzimat era's reform movements highlighted the belief that progress could be achieved only through science, prompting the establishment of a Darülfünun (University) on July 23, 1846, through a decree. Although the first attempt at Darülfünun faltered, a renewed effort in 1870 resulted in Darülfünun-u Osman's establishment, aiming to align with modern scientific ideals. Yet, this venture faced setbacks as well. Eventually, in 1900 during Sultan II. Abdülhamit's reign, Darülfünun-u Şahane emerged, which laid the foundation for consistent university education and witnessed the introduction of various disciplines.

Following the declaration of the Republic, Istanbul University was founded on August 1, 1933, as a successor to the earlier Darülfünun. The university's growth over nearly 90 years has cultivated numerous well-trained professionals across a spectrum of fields. Istanbul University-Cerrahpaşa came into existence in 2018, encompassing various faculties and units that branched out from Istanbul University. The current Istanbul University-Cerrahpaşa hosts thousands of students, educators, and personnel across its campuses, contributing significantly to education and research.
